Protein Purification Tray (3P)

This technical platform brings together expertise in the purification of proteins and antibodies directed against the components of food products and their raw materials (proteins, polysaccharides, phenolic compounds).

Thanks to its equipment and expertise, the platform is able to purify and characterise proteins of plant, animal and recombinant origin as part of collaborative projects or for independent use by academic laboratories and private companies.

These purified proteins are used to study structure-function relationships, whether in terms of techno-functional properties or enzymatic or biological activities.

The platform also uses the Transform department's collection of antibodies dedicated to foods and their raw materials.

Various substrates are processed:

  • Cereal, legume and oilseed proteins
  • Food proteins of animal origin (milk, egg)
  • Biologically active proteins (enzymes and enzyme inhibitors).
  • Recombinant proteins (antibodies, allergens, etc.)

Technology and expertise

The aim of the platform is to purify proteins from natural or heterologously expressed raw materials.

  • The platform develops and implements extraction and separation methods for the preparation of protein-enriched fractions down to the preparative scale (10 mg-10 g): centrifugation, ultrafiltration, low-pressure chromatography.
  • The platform is involved in characterising enriched extracts and purified proteins: protein content, composition, yield, purity, conformational state, activity, etc.
  • The platform is equipped with flow-force fractionation (FFF) equipment coupled to a light-scattering detector, which enables the size and size distribution of polymers or nano-objects (1 nm to 1 μm) to be characterised.

Accessibility

The platform is accessible to all members of the unit and external collaborators, with the agreement of the platform manager, for independent use of the equipment (after training) or for the development of collaborative projects.

Proteins are purified according to specifications defined with the project leader.

Users of the platform fund the purchase of consumables required to carry out their project, and the maintenance of the equipment in the form of a contribution.
